Not much rest. They normally play about 6 - 7 games in a row, then a day off - rinse and repeat for 6 months.
Unlike other sports the actual game itself isn't as exhausting (lot of standing around, waiting for some action to come their way), but the wear and tear of a long season, with not much down time does get extremely tiring.
Pitchers normally only pitch once every 6 games or so. (depends on how many different pitchers the team has in it's rotation). Also most pitchers usually pitch into the 6th or 7th inning (depending on "pitch count"), then a relief pitcher / closer will take over to finish the game.
Probably the hardest job is that of the catcher. Day after day of squatting down, catching balls being thrown at them over 90 mph (I can't imagine the wear and tear on their knees).
The first baseman also gets a lot of action. Sometimes an outfielder might only get the ball hit to them a couple times a game.
Most players get about 4 times at bat (again, depends on the score as to how often they come to the plate).
Games take around 3 hours to play.
